How to prepare samples for better analysis results?

Dear Reader,

Does your sample matrix contain high molecular weight substances that you want to separate from your analytes because they would interfere with your analysis process? For example, the analysis of pesticides in edible oils is one such application. In that case, gel permeation chromatography (GPC), which separates the components of a mixture according to their size without any chemical interaction, may be one of your routine tasks.

Are you still conducting your GPC process manually?

If you do not mind the manual GPC cleanup process and you do not find it time-consuming, do not read on. If you continue reading here, you will learn how to improve sample cleanup and gain time.

Automated GPC sample preparation improves quality and saves time

The AZURA® GPC Cleanup System automates labor-intensive and time-consuming cleaning tasks based on GPC. Never again miss the desired fraction, but instead collect your product exactly according to the detector signal. Reproducibility and quality of sample processing are thereby improved.

Since the system can process up to 15 samples fully automatically after loading, you save time. Based on the versatile AZURA® device platform, individual customer requirements can be met. The cost-effective Mobile Control software offers a touch-optimized user interface for intuitive control of the devices without a PC.

Application
Application: GPC Cleanup of olive oil samples
GPC cleanup of olive oil samples

This application describes a sample preparation method for the analysis of pesticide residues in olive oil.

Pesticides were separated from the oil matrix by size exclusion/gel permeation chromatography (GPC) according to US EPA SW-846 method 3640A.

The optimized GPC purification technique was carried out with a KNAUER AZURA GPC Cleanup System for automated sample cleaning.

Basics
How does cleanup with size exclusion or gel permeation chromatography work?

GPC Cleanup (gel permeation chromatography) can be employed for performing general cleanup tasks on a wide range of sample matrices such as foods, tissues, plants, and environmental samples. The separation of components takes place according to their molecular size and shape due to the heterogeneous pore size of the solid phase (illustration below). Somewhat surprising, perhaps, is that large molecules elute first.

How can this be explained? Smaller molecules are better able to penetrate the pores of the stationary phase. Thus, large molecules, which are less able to enter the pores of the stationary phase move more quickly with the mobile phase and elute earlier.

With this method high-molecular substances such as lipids and proteins, potentially interfering with subsequent analysis e.g. of pesticides, can be efficiently removed.
